摘要
我们利用ABC—ELISA方法检测抗胃癌单克隆抗体,并与常规ELISA方法进行比较。ABC—ELISA方法的敏感性比常规ELISA方法高110～100倍,但非特异性并没有增加。重复5次检测杂交瘤上清液的效价,结果基本一致。本文所有结果表明,ABC—ELISA方法用于筛选和检测抗胃癌单克隆抗体不但灵敏度高,而且特异性和重复性好,是值得作为筛选和检测单克隆抗体的新方法而进行推广。
The authors obtained satisfactory results in using ABC-ELISA to screen McAbs against gastric cancer and comparing this method with the standard ELISA. Though the former was 10-100 times more sensitive than the latter,non-specificity was not obvious. The results of measurement of the titer of hybridoma supernatant repeated for five times were almost similar. These results indicated that ABC-ELISA used in assaying surface antigens is not only highly sensitive,but also strongly specific, with property of repetition satisfactory. It is valuable to be popularized as a method for screening and measuring McAb agsinst surface antigens of tumors.
