摘要
目的探讨吲哚胺2,3-二氧化酶(IDO)在前房相关免疫偏离(ACAID)形成中的作用。方法分别将卯白蛋白(OVA)及其与完全弗氏佐剂的混合液注入BALB/C小鼠前房及后足垫内,皮内注射OVA观察迟发型超敏反应,评价ACAID的形成。分别于小鼠前房注射OVA后第3、7、10和14d处死动物,取其脾脏,采用免疫组织化学和Westernblot法检测小鼠脾脏组织中IDO的表达及其动态变化。结果正常小鼠脾脏组织中有少量IDO表达;前房注射OVA后第7、10和14dIDO表达量明显增加(P<0.05)。免疫组织化学显示的阳性细胞数量变化与Western-blot检测的蛋白动态变化相一致。结论在ACAID形成过程中IDO表达显著性增加,提示它可能参与了ACAID的形成。
Objective To observe the role of Indoleamine 2 ,3-dioxygenase( IDO) in anterior chamber associated immune deviation (ACAID) induced by ovalbumin (OVA) in BALB/c mice. Methods A dosage of 100 μg OVA(20 mg/mL) was injected into the anterior chamber (AC) of BALB/c mice to induce ACAID,and followed by an immunization with OVA 250 μg OVA(100 μL) combined with complete Freund on day 3,7,10 and 14 after AC injection. Delayed-type hypersensitivity (DTH) response was evaluated 7 days after the immunization. Immunohistochemistry and Western blot were performed to determine the dynamic changes and expression of IDO protein respectively at each time point. Results A significant DTH reaction was induced 8 days after subcutaneous injection of OVA in the control group. An impaired DTH reaction was observed on day 15 ,18 and 22 respectively after AC injection in 7-, 10- and 14-day groups,respectively. IDO was weakly detectable in the normal spleen and increased significantly at 7 ,10 and 14 days after AC inoculation. Conclusion An increased expression of IDO during the formation of ACAID indicates that it involves in the development of this immune deviation.
