特发性血小板减少性紫癜患者糖皮质激素受体的表达 被引量:2

Expression of glucocorticoid receptors in steroid-resistant patients with idiopathic thrombocytopenia purpura

摘要 目的探讨特发性血小板减少性紫癜(ITP)患者糖皮质激素治疗效应与外周血单个核细胞(PBMC)内两种糖皮质激素受体(GR)亚型-GRα和GRβ,及C-FOX表达水平之间的关系。方法用逆转录-聚合酶链反应(RT-PCR),检测激素敏感型、激素抵抗型ITP患者和正常对照组GR,αGRβ,C-FOX的mRNA的表达。结果ITP患者和正常对照组均有GRα、GRβ,C-FOSmRNA的表达;激素抵抗型ITP患者GRβmRNA表达明显高于激素敏感型ITP患者及正常对照组(P<0.05);GRα,C-FOXmRNA在各组无显著差别。结论ITP患者糖皮质激素抵抗可能与GRβ的表达亢进有关。 Objective To investigate the relationship between the response to glucocor ticoids and glucocorticoid receptors(GR) isform α and β expression or C-FOS expression in PBMC with idiopathic thrombocytopenia purpura(ITP). ([WT10.HZ]Methods[WT] The) RT-PCR was used to detect the expression of GRαGRβ and C-FOXs in PBMC with ITP patients with sensitive or resistant response to glucocorticoid and controls. Results[WT] The expression of GRα mRNAGRβ mRNA and C-FOX mRNA was detected in all patients and controls. The expression of GRβ mRNA in the resistant group was significantly higher than that in the sensitive group and controls(P<0.05 ). The expression of GRα mRNA and C-FOX mRNA was not different in all groups. ConclusionWT The results suggest that glucocorticoid resistance in idiopathic thrombocytopenia purpura may be related with the over-expression of GRβ.
