机采血小板低温保存的研究与观察

摘要 目的提高血小板深低温保存的质量及临床应用观察。方法采集的血小板悬液加入DMSO5%后置于-80℃的深低温冰箱,复融中观察蛋白析出率及冻存血小板的回收率,黏附和聚集功能,第3因子活性的测定。结果-80℃深低温保存1a内的血小板在复融中蛋白析出率明显降低,血小板回收率≥70%,黏附聚集功能和第3因子活性完好。结论DMSO终浓度5%的冻存血小板在1a内各项主要功能指标在正常范围内,临床应用效果好。 Objective To improve the deep low temperature storing quality of blood platelet and their clinical applications.Methods 5% DMSO was added into the collected blood platelet suspensions,then put them into the refridgerator below-80 ℃;examine the volume of the protein separated out from the suspension during remelting and the recycling rate for the blood platelet, the functions of agglomeration and adherence,also the activity of the cytokine 3'.Results The falling rate of the protein reduced apparently when remelted the blood platelet stored at-80 ℃ temperature within 1 year, and the recycling rate of the blood platelet ≥70%,good agglomeration and adherence properties and cytokine 3' activities indicated. Conclusion The freezing storing blood platelet in the 5% DMSO solution shows positive results for the clinical applications and all the major criterions are under the requirements even after 1 year.
