二氧化碳气腹对人卵巢癌细胞株SKOV_3VEG F和MMP9表达的影响 被引量:13

摘要 目的探讨腹腔镜二氧化碳(CO2)气腹对恶性卵巢肿瘤细胞侵袭能力的影响。方法体外培养人卵巢浆液性乳头状腺癌SKOV3细胞株,以CO2处理2 h,应用细胞爬片免疫组织化学法和半定量RT-PCR方法检测不同时相中血管内皮生长因子VEGF和MMP9表达水平。结果CO2处理后12 h SKOV3细胞VEGF和MMP9蛋白和mRNA表达显著增加,VEGF在CO2处理后24 h表达达到高峰,MMP9在48 h达到高峰。结论CO2可以增强卵巢癌细胞SKOV3的侵袭能力。 Objective To study the effects of carbon dioxide on the metastatic capability of ovarian carcinoma cell line. Methods After ovarian carcinoma cells SKOV3 were treated with 100% carbon dioxide for 2 h, the expressions of VEGF and MMP9 were examined by immunohistochemistry and RT-PCR. Results After the treatment of carbon dioxide for 2 h, the expression of VEGF and MMP9 was significantly increased at 12^th h, the expression of VEGF reached the peak at 24^th h and the expression of MMP9 reached the peak at 48^th h. Conclusion Carbon dioxide may improve the metastatic capability of ovarian carcinoma cell line.
