表达人KIAA1173基因的鼻咽癌6-10B细胞模型的建立

摘要 目的建立表达人KIAA1173基因的鼻咽癌6-10B细胞模型,为探究人KIAA1173基因在鼻咽癌发病机制中的作用奠定实验基础。方法从新鲜肌肉组织中提取总RNA,采取逆转录PCR(RT-PCR)得到人KI-AA1173基因cDNA,纯化PCR产物,与真核表达载体pcDNA3.1(+)连接,转化,铺板,筛选阳性克隆,酶切鉴定并进行序列测定。将重组质粒pcDNA3.1(+)-KIAA1173和空载体利用阳离子脂质体介导转入鼻咽癌6-10B细胞,经G418筛选阳性克隆。用RT-PCR、原位杂交及免疫细胞化学方法检测KIAA1173基因在6-10B细胞中的稳定表达情况。以空白质粒转染组作为对照。结果重组质粒pcDNA3.1(+)-KIAA1173经酶切和DNA测序分析显示与GenBank所公布的相应序列相符,插入方向正确,阅读框架完整。转染KIAA1173基因的6-10B细胞阳性克隆在mRNA和蛋白水平证实,转染细胞内有该基因高表达,表明细胞可表达人KIAA1173基因。结论成功获得了人KIAA1173基因全长cDNA的基因克隆,将该基因导入鼻咽癌6-10B细胞后,获得生物学活性稳定的KIAA1173基因高表达的细胞模型。
