蜕膜细胞条件培养液中TNF-α及EGF对卵巢癌细胞株COC1侵袭基因MMPs/TIMPs表达的影响 被引量:1

摘要 目的研究蜕膜细胞条件培养液(decidual conditioned media,DCM)中TNF-α及EGF对卵巢癌细胞株COC1侵袭基因基质金属蛋白酶(MMPs)/基质金属蛋白酶抑制剂(TIMPs)表达的影响。方法原代培养早孕蜕膜细胞并提取DCM,用DCM及分别加入TNF-α及EGF抗体的DCM处理COC1,利用RT-PCR法对COC1侵袭基因MMPs/TTMPs的表达进行分析。结果COC1表达MMP-2,TIMP-2,而不表达MMP-9,TIMP-1。早孕EGF-Ab组与对照组(DCM组)比较,前者可以使COC1的MMP-2 mRNA表达降低,且差异有显著性(P<0.01)。而TNF-α抗体组可以降低MMP-2 mRNA的表达,但与对照组比较差异无显著性(P=0.280)。与DCM组比较,EGF-Ab组可以使COC1的TIMP-2 mRNA表达增加,两者差异有显著性(P<0.01)。而TNF-α抗体组有增加TIMP-2 mRNA表达,但差异无显著性(P>0.05)。结论早孕DCM中TNF-α可能抑制COC1侵袭力。早孕DCM中EGF可能促进COC1侵袭力。
