三氧化二砷联合全反式维甲酸对小鼠肝癌细胞H22及移植瘤的抑制作用 被引量:3

摘要 目的探讨三氧化二砷(arsenic trioxide,As2O3)联合全反式维甲酸(all trans retinoic acid,ATRA)在体内和体外对小鼠肝癌H22细胞的抑制作用。方法建立小鼠肝癌H22移植瘤模型,观察As2O3联合ATRA的体内抑瘤作用;用四甲基偶氮唑蓝(MTT)方法测定As2O3联合ATRA在体外对小鼠肝癌H22细胞株的生长抑制作用。结果As2O3联合ATRA在体外能明显抑制H22细胞的生长和增殖,抑制率为52.24%;在体内对H22小鼠的肿瘤体积、重量均有明显的抑制作用,抑制率分别为66.25%、62.07%。结论As2O3和ATRA在体内和体外对H22肿瘤细胞的生长均有抑制作用,联合应用作用加强,有协同作用。 Objective To study the inhibition effects of arsenic trioxide(As2 O3 )and ATRA (all trans retinoic acid) on murine hepatocarcinoma cell H22 in vitro and in vivo .Methods The transplant tumor mouse model was used to observe the inhibition effects of As2 O3 and ATRA on H22 cells /n vivo. Using MTT assay to determine the inhibition effect of As2O3 combined ATRA on the grouth of cultured H22 cells was determined by MTT assay in vitro. Results The combination of As2 O3 and ATRA could obviously inhibit the growth and proliferation of cultured H22 cells. The inhibition rates were 52.24%. It also had significant anti-tumor activity in transplant hepatoma mice model. The inhibition rates were 66.25 % (size) and 62.07 % (weight). Conclusion As2 O3 and ATRA have marked inhibition effects on H22 hepatocarcinoma cells in mice both in vitro and in vivo. The enhanced activity might result from the combination of As2 O3 and ATRA.
