抗白介素-9抗体对BALB/c哮喘小鼠T细胞功能的影响

摘要 目的通过比较不同干预措施对小鼠哮喘模型的影响情况,研究其对免疫系统中CD4+T辅助细胞2型(Th2细胞)活性的影响,旨在了解外源性抗白介素(IL)-9抗体(Anti-IL-9 Ab)在哮喘中的潜在治疗价值。方法 30只BALB/c小鼠随机均分为阳性对照组、Anti-IL-9 Ab组和地塞米松(Dex)组,建立小鼠哮喘模型,采取不同干预措施。比较各组血清、脾和骨髓细胞培养上清液细胞因子的含量。结果 Anti-IL-9 Ab组小鼠血清中 TH2细胞因子较阳性对照组明显降低(P<0.01),Dex组小鼠血清和脾细胞培养上清液中Th2、Th1细胞因子均较阳性对照组明显降低(P<0.01),两组骨髓细胞培养上清液细胞因子的含量较阳性对照组均无明显变化。结论 Anti-IL-9 Ab和Dex均能明显减轻哮喘小鼠的肺部炎症,在气道建立起新的Th1/Th2平衡。与Dex相比,应用Anti-IL-9 Ab建立的新平衡不会造成因细胞免疫功能受抑引起的一系列不良反应。 Objective To investigate the therapeutic potential of interleukin (IL)-9 blockage in asthma and to determine the influences of anti-IL-9 antibody on the functions of Th2-type CD^4+ T cells of asthmatic mouse model with different interventions. Methods 30 BALB/c mice were divided randomly into 3 groups, positive controlled group, Anti-IL-9 Ab group and Dex group. Different interventions were conducted after construction of asthmatic mouse model. Cytokines in the serum, supernatant of splenic cell or bone marrow cell culture were evaluated. Results Anti-IL-9 antibody decreased the level of Th2 cytokine in serum, but dexamethasone decreased the level of Th1 and Th2 cytokines in both of the serum and the supernatant of splenic cell culture. The cytokines in the supernatant of bone marrow cell culture did not change significantly. Conclusion Both anti-IL-9 antibody and dexamethasone can alleviate the pulmonary inflammation of asthmtic mouse, a new balance of Th1/Th2 can be set up within the airway. In contrary to dexamethasone, the new balance set up by anti-IL-9 antibody would not cause serial adverse reactions by inhibiting the cellular immunity.
