核因子κB及其抑制蛋白IκBα在成年大鼠坐骨神经许旺细胞的分布 被引量:1

摘要 目的:观察核因子κB及其抑制蛋白IκBα在成年大鼠坐骨神经许旺细胞的表达,并分析二者之间的关系及意义。方法:实验于2005-01/06在重庆工学院生物工程学院及第三军医大学大坪医院野战外科研究所完成。随机取成年雄性SD大鼠6只,体质量200~250g。1%戊巴比妥钠(20~40mg/kg)腹腔麻醉后打开胸腔,用4%多聚甲醛溶液经左心室灌注后,取左侧坐骨神经,采用p65和p50(由p65和p50两个亚基组成的异源二聚体在核因子κB组成中发挥主要功能)及抑制蛋白IκBα的多克隆抗体进行免疫组织化学染色,检测核因子κB及其抑制蛋白IκBα在正常成年大鼠坐骨神经许旺细胞的表达及分布。结果:①p65和p50在坐骨神经许旺细胞胞浆中表达均很弱,胞核中则不表达。②IκBα则在胞浆中有较强的表达,胞核中不表达。结论:核因子κB及其抑制蛋白IκBα在正常成年大鼠坐骨神许旺细胞经均有一定程度的表达,在胞浆中前者表达较弱,后者较强,二者在胞核中均不表达。 AIM: To observe the expression of nuclear factor-kappa B (NF-kB) and its profilin I kappa B alpha (IkBα) in sciatic nerve sehwann cells (SCs) of the adult rat, and analyze the relationship and meaning of the two. METHODS: This experiment was carried out in the College of Bioengineering, Chongqing Institute of Technology and Research Institute of Surgery, Daping Hospital, Third Military Medical University of Chinese PLA from January to June 2005. Six male adult Sprague-Dawley rats, with body mass of 200 to 250 g, were anesthetized with 1% Barbiturate natrium solution (20-40 mg/kg), thoracic cavities were opened and 4% paraformaldehyde was infused from left ventricle, then left sciatic nerves were harvested and immunohistochemical stain was applied with p65, p50 (Heterodimer constituted by p65 and pS0 two subunits exerted major function in the NF-kB) and IkBα rabbit polyclonal antibodies. Expression and distribution of NF-kB and IkBα were detected in the sciatic nerve sehwann cells. RESULTS: Protein expression level of p65 and p50 were very low in the cytoplasm of sciatic nerve SCs and no expression appeared in nucleus, however, the level of IkBα was very high in cytoplasm, while no expression was found in nucleus either. CONCLUSION: NF-KB and IkBα could express in sciatic nerve SCs of the adult rat to some extent, the former expressed lowly while the latter highly in cytoplasm, neither expressed in nucleus.
