摘要 目的:用 HPLC 梯度洗脱法同时测定蛇胆中牛磺胆酸(TCA)、牛磺鹅去氧胆酸(TCDCA)、牛磺去氧胆酸(TDCA)、甘氨胆酸(GCA)4种结合型胆汁酸的含量。方法:用美国 Zorbax Extend C_(18)柱,乙腈-0.15%磷酸氢二钠溶液梯度洗脱0~20min(15:85~40:60)。流速1.0mL·min^(-1),检测波长203nm。结果:TCA、TCDCA、TDCA、GCA 在进样量为2~39μg范围内线性关系良好。该方法回收率 TCA 为99.5%(RSD=0.5%),TCDCA 为100.3%(RSD=1.8%),TDCA 为100.0%(RSD=1.8%),GCA 为98.9%(RSD=2.2%)。结论:HPLC 梯度法能将蛇胆中结合型胆汁酸很好分离,方法简单,准确。 Objective: An HPLC gradient elution was studied for the determination of TCA , TCDCA, TDCA, GCA in snake gall Methods:C18 column was used with a mobile phase of acetontrile -0. 05% Na2HPO4 ,0 -20 min of ( 15:80 -40: 60) gradient elution post time was 20 min,the wavelenth of detected was set at 203 nm. Results:TCA , TCDCA, TDCA , GCA calibration curves were linear at the range of 2 - 39 μg. The average recoveryies rate TCA was 99. 5% ( RSD =0. 5% ) ,TCDCA was 100. 3% ( RSD = 1.8% ) ,TDCA was 100. 0% ( RSD = 1.8% ) ,GCA was 98. 9% (RSD = 2. 2% )respectively. Conclusion: The method is simple and accurate. The results show that the method can be used to control quality of Snake gall.
