摘要
目的:为锁阳药材最佳采药期的确定提供科学依据。方法:用高效液相色谱法,对不同地区、不同生育期天然锁阳与人工栽培锁阳的儿茶素含量的动态变化进行了研究。色谱条件:色谱柱为 ZORBAX SB-C_(18)柱(4.6mm×250mm,5μm),0.04mol·L^(-1)枸橼酸溶液-N,N 二甲基甲酰胺-四氢呋喃(50:4:1)为流动相,流速1.0mL·min^(-1),检测波长为280nm.柱温:35℃;进样量:30μL。结果:无论不同地区天然的还是人工栽培锁阳不同生育期儿茶素含量均呈现规律性变化,出土前期的儿茶素含量最高,出土期的次之,结实期的最低。结论:经研究确认儿茶素为锁阳药材的有效成分之一,也可作为其指标性成分之一。如仅以儿茶素作为指标性成分,出土前期和出土期所采集的锁阳药材质量最好。该结果同锁阳药材的民间采药期-即5月初出土期采药是吻合的,为锁阳药材采收期的确定和质量评价提供了科学依据。
Objective:To provide scientific foundation for establishing collecting - time of best quality of herb. Methods:In this paper catechin was determined by HPLC to study dynamic trends of catechin content of the wild and cultivated Cynomorium songaricum Rupr. in different growth stages and different areas. A ZORBAX SB - C18 column(4.6 mm ×250 mm,5 μm) was used. The mobile phase consisted of 0.04 mol · L^-1 citric acid solution- N,N- dimethylformamide- tetrahydrofuran(50: 4:1 ) at a flow rate of 1.0 mL · min^-1. Detection wavelength was set at 280 nm and column temperature was 35 ℃. The injection volume was 30 μL. Results:The catechin content of Cynomorium songaricum Rupr. changes regularly. The catechin content of Cynomorium songaricum Rupr. that of is higher at proenearth stage than that of unearthed. The content at unearthed stage is higher than that in flowering stage. In flowering stage is higher than fruits developing stage. Conclution : Catechin has been regarded as one of active components and one of mark components. When the catechin was selected as the mark component, the quality of Herba Cynomorii that is proenearth stage or the unearthed was best. This result accorded with folk - custom by which Herba Cynomorii was usually collected in the beginning of May. Our research provided a scientific foundation for the right collecting - time and the quality evaluation of Herba Cynomorii.
