糖尿病大鼠缺血再灌注脑组织核转录因子-κB的表达及意义 被引量:1

Expression of NF-κBp65 and the Clinical Significance in Diabetic Rats model with Cerebral Ischemic Reperfusion

摘要 目的观察糖尿病大鼠缺血再灌注脑组织NF-κBp65的表达及意义。方法89只Wistar大鼠随机分为正常对照组、假手术组、正常血糖缺血再灌注组(nIR)和糖尿病缺血再灌注组(dIR),观察时间点为再灌注0小时、6小时、12小时、24小时,每个时间点各5只。用小剂量链脲佐菌素(STZ)腹腔注射诱发形成糖尿病大鼠模型,采用线拴法复制大鼠大脑中动脉缺血再灌注模型,应用免疫组化的方法测定NF-κBp65的表达。结果dIR组和nIR组NF-κBp65主要表达于缺血周边区,dIR组再灌注0小时、6小时、12小时、24小时NF-κBp65阳性细胞百分率分别为24.7±4.2、53.4±8.0、72.4±7.2、60.7±5.0,与nIR组同一时间点比较,差异有显著性(P<0.05)。结论糖尿病大鼠缺血再灌注脑组织NF-κBp65表达增加和表达时间提前可能是糖尿病加重脑缺血再灌注损伤的机制之一。 Objective To investigate the expression of NF-κB, and the clinical significance in diabetic rats model with cerebral ischemic reperfusion. Methods 89 wistar rats were randomly divided into four groups; normal group, NS control group, normoglycemic cerebral ischemic reperfusion group (nIR) and diabetic cerebral ischemic reperfusion group (dIR). Diabetic animal model were made by injection of streptozocin (STZ) through abdomen. The rats model of cerebral ischemia reperfusion were developed by thread embolism method, and reperfusion for 0h, 6h, 12h and 24h(n=5). The expression of NF-κb were detected by irnmunohistochernistry. Results Expression of NF-κBp65 increased in dIR group, which were 24.7±4.2, 53.4±7. 9,72. 4±7. 2,60. 7±5.0(%) in reperfusion for 0h, 6h,12h, 24h respectively, compared with the same time in nlR group (P〈0. 05). Conclusion NF-κB may play an important roles with local cerebral ischemic reperfusion in diabetic model rats.
