摘要
目的探讨肾脏水通道蛋白2(AQP2)的表达与糖尿病肾病大鼠水液代谢异常及泽黄煎剂防治作用的机制。方法用高糖高脂饲料刺激加小剂量STZ复制糖尿病大鼠模型,随机分为模型组、阳性对照组、泽黄煎组,另设正常组。免疫组织化学检测AQP2在各组大鼠肾脏中的分布及蛋白质的表达;RT-PCR方法检测各组大鼠AQP2mRNA的表达,并以β-actin相对定量。结果泽黄煎组和阳性对照组空腹血糖、24h尿蛋白排泄率较模型组均显著下降。肾免疫组化显示:AQP2主要在肾脏的集合管表达。与模型组比较,泽黄煎组动物的肾脏AQP2蛋白、AQP2mRNA的表达均明显下调(P<0.05)。结论模型组大鼠肾组织中AQP2表达增多,AQP2增多可能参与了糖尿病多尿的形成。泽黄煎剂对水通道蛋白2的表达有下调作用,这可能是其调节水液代谢作用的机制,也是其防治疗糖尿病肾病的基础。
Objective To observe the relationship between expression of aquaporin protein2(AQP2) and abnormal fluid metabolism in diabetic nephropathy (DN) rats and to explore the therapeutic mechanism of Ze - Huang Decoction (ZHD). Methods Diabetic rat model was induced by high glucose and high fat feedstuff and low - dosage streptozotocin. The rats were randomized into model group, positive control group, ZHD group, and a normal control group was also set up. Immuno - histochemical method was adopted to detect the distribution of AQP2 in rats kidney as well as the expression of proteins. RT - PCR method was employed to detect the expression of AQP2 mRNA in rats, with β - actin as relative quantification. Results The blood glucose and 24 - hour urine protein levels in ZHD group and positive control group obviously came clown as compared with that in the model group. Immuno - histochemical results indicated that AQP2 mainly expressed in the collecting tube of kidney. Compared with the ZHD group were both lowered (P〈0.05). Conclusion The rat model, and the increase might participate in the formation sion of AQP2, which may be its mechanism of regulating fluid DN. model group, the expressions of AQP2 and AQP2 mRNA in expression of AQP2 increases in the renal tissue of diabetes of diabetes polyuria. ZHD can down - regulate the expres-metabolism, and also be the basis of preventing and treating DN.
