摘要
以唇形科鞘蕊花属彩叶草(Coleus blumei Benth)叶片为外植体,进行组织快繁的研究。结果表明:叶片组织培养中,最适宜的消毒处理为消毒剂采用0.1%升汞溶液,加入3~4滴吐温80,消毒时间为5min,总体污染率低于20%;最适的愈伤组织诱导及继代培养基为1/2MS+2.0mg/L6-BA+1.0mg/LNAA的组合,其平均诱导率为72%;最适分化培养基是1/2 MS+1.0mg/L 6-BA+0.5mg/LNAA的组合,其平均分化率为85.4%;彩叶草不定芽的最适生根培养基是1/2MS+0.5mg/LNAA组合,其平均生根率为92.5%。
The study of the plant tissue culture constructed rapid propagation system of Coleus blumei Benth. with its leaves as the explants. From this research, the main results were as follows: In the investigation of plant tissue culture with leaves of Coleus blumei Benth. , the optimum disinfection method was using 0. 1% solution of mercuric chloride as the disinfectant with 3-4 drop of Tween-80, and the disinfection last 5 min. The rate of pollution with this method was below 20%. The optimum culture medium of inducement callus and sub culture was 1/2MS+2. 0 rng/L 6-BA+1.0 rng/L NAA, and its average rate of inducement was 72%. The optimum culture medium of differentiation culture was 1/2MS+1.0 mg/L 6-BA+0. 5 rng/L NAA, and its average rate of differentiation was 85. 4%. The optimum culture medium of rooting culture was 1/2MS+0. 5 mg/L NAA,and its average rate of rooting was 92. 5%.
