摘要
目的观察槲皮素对胃癌细胞SGC-7901生长及HSP70和EGFR的影响。方法以台盼蓝拒染法计数胃癌细胞的生长抑制率,透射电镜从形态变化上了解凋亡的发生,流式细胞术检测细胞周期变化,免疫组化检测HSP70和EGFR的表达。结果台盼蓝拒染法计数显示槲皮素抑制SGC-7901细胞增殖的作用明显,且呈浓度和时间依赖性,槲皮素处理48 h后的IC50为14.12μmol/L。形态学检测显示出细胞凋亡的特征变化,流式细胞仪检测表明经10-20μmol/L的槲皮素处理,SGC-7901细胞周期阻滞于G0/G1期,经10μmol/L槲皮素处理的SGC-7901下调HSP70和EGFR蛋白的表达。结论槲皮素能抑制胃癌细胞的生长,呈时间剂量依赖性,能诱导SGC-7901发生凋亡,使细胞周期阻滞于G0/G1期,抑制增生与诱导凋亡的机制可能与下调HSP70和EGFR的表达有关。
To investigate the effects of quercetin on human gastric cancer lines SGC-7901 and on the expression of heat shock protein 70 (HSP70) and epidermal growth factor receptor (EGFR). Methods The number of viable cells was ascertained by trypan blue dye exclusion test. Morphological changes of apoptotic cells were observed by electronic microscopy. Cell cycle was analyzed by flow cytometry. HSP70 and EGFR expression was assayed by immunocytochemistry. Results Quercetin had a significant inhibition on the growth and proliferation of human gastric cancer line SGC-7901 in vitro in a concentration-dependent manner. The IC50 was 14.12/μmol/L after exposure to quercetin for 48 h. Electronic microscopy provided evidence that apoptosis occurred in the gastric cancer cells of treatment groups. FCM analysis showed that quercetin arrested SGC-7901 cells at the G0/G1 phase. The immunocytochemical study showed that the expression of HSP70 and EGFR was significantly inhibited by 10/μmol/L quercetin. Conclusion Dose-dependent inhibitory effects of quercetin on the growth of human gastric cancer cell line are remarkable. Quercetin can induce apoptosis of SGC7901 and keep the cell cycle at G0/G1 phase from down-regulating expression of HSP70 and EGFR.
