生脉解毒通络胶囊对糖尿病大鼠心肌组织ECM积聚的影响 被引量:3

摘要 目的:研究糖尿病大鼠心肌组织ECM积聚及中药生脉解毒通络胶囊的调节作用。方法:46只Wistar大鼠用链脲佐菌素诱发糖尿病后,随机分为DM组、苯那普利治疗组和生脉解毒通络胶囊治疗组,以正常10只大鼠作为对照。连续处理12周后,应用免疫组化方法检测心肌组织的基质金属蛋白酶(MMP-1)及其抑制物(TIMP-1)和Ⅰ、Ⅲ型胶原的蛋白表达水平。结果:DM大鼠TIMP-1及Ⅰ、Ⅲ型胶原蛋白表达增加,MMP-1表达减少,生脉解毒通络胶囊组心肌组织TIMP-1及Ⅰ、Ⅲ型胶原蛋白表达明显低于DM组(P<0.01),MMP-1蛋白表达明显高于DM组(P<0.01)。结论:生脉解毒通络胶囊对DM心肌病变有保护作用,其机制可能是通过改善糖脂代谢,减少细胞外基质的积聚。 Objective: To develop the Myocardial ECM Accumulation of Diabetes Rats and the mechanisms of Shengmai Jiedu Tongluo Capsule. Methods : 46 Wistar rats were evoked to DM model with Streptozotocin and randomized to 4 groups: DM group, Benazepril group, Shengmai Jiedu Tongluo Capsule group and normal control group. After 12 weeks,the Expression of MMP - 1 ,TIMP - 1 and Collagen Ⅰ,Ⅲwere detected with the methods of immunohistochemical staining. Results: The expression of TIMP - 1 and Collagen Ⅰ,Ⅲ of DM rats raised and the expression of MMP - 1lowed. With the comparison of DM group, the expression of TIMP - 1 and Collagen I , Ill ofShengmai Jiedu Tongluo Cap- sule group lowed obviously( P 〈0.01 )and the expression of MMP- 1 ofShengmai Jiedu Tongluo Capsule group raised obviously( P 〈0. O1 ). Conclusion: Shengmai Jiedu Tongluo Capsule protect DM myocardial. The mechanisms maybe improve glycolipid metabolism and reduce extracellular matrix accumulation.
