不同试剂检测北京地区健康献血者淋巴细胞CD3^+CD4^+、CD3^+CD8^+百分比参考值调查 被引量:1

摘要 目的探讨北京地区健康献血者CD3+CD4+、CD3+CD8+百分比的参考值范围,并对国产和进口的淋巴细胞免疫表型双染试剂进行比较。方法收集北京地区500例健康献血者全血标本,分别使用进口和国产的CD3FITC/CD4PE和CD3FITC/CD8PE双染试剂,应用同一台流式细胞仪进行检测,并对不同性别、民族和年龄段的结果以及不同试剂的结果进行比较。结果进口试剂检测的北京地区健康献血者CD3+CD4+、CD3+CD8+百分比以及CD3+CD4+/CD3+CD8+的范围(95%可信限)分别为22.92~49.96、14.86~41.70、0.37~2.47,不同年龄段(18~29、30~39与40岁以上组)各项指标均值的差异有统计学意义(P<0.05),而不同性别的比较仅CD3+CD4+(%)的差异有统计学意义(P<0.01),比较汉族(n=484)与少数民族(n=16)中各项指标的差异均无统计学意义(P>0.05)。国产与进口试剂的检测结果显著相关,CD3+CD4+(%)、CD3+CD8+(%)以及CD3+CD4+/CD3+CD8+的r值分别为0.970、0.953、0.967。u检验结果显示CD3+CD4+(%)的差异无统计学意义(P>0.05),而CD3+CD8+(%)、CD3+CD4+/CD3+CD8+的差异有统计学意义(P<0.01)。结论初步建立北京地区健康献血者CD3+CD4+(%)、CD3+CD8+(%)的参考值,参考值受年龄、性别、民族、检测试剂和仪器等多种因素的影响。国产和进口淋巴细胞免疫表型双染试剂检测结果基本一致,相关性具有显著性。 Objective To study reference value of CD3^+CD4^+ percentage,CD3^+CD8^+ percentage in healthy blood donors in Beijing and to compare results among imported and domestic dual-dye T lymphocyte immunophocyte reagents. Methods 500 whole blood specimens from healthy blood donors in Beijing were collected and measured by flow cytometer using imported and domestic reagents respectively. The differences of the reference value divided by age, gender, ethnic and reagent were analyzed. Results The reference value of CD3^+CD4^+ percentage,CD3^+CD8^+ percentage and CD3^+CD4^+ vs. CD3^+ CD8^+ratio (at 95 %CI) were 22.92- 49.96, 14.86- 41.70, 0. 37- 2. 47 measured by imported reagents. There were statistically significances (P〈0. 05) for results from different age groups (18-, 30- compared with the group above 4 0). Only CD3^+CD4^+ percentage between male and female had statistically significances (P〈0.01) and no differences (P〉0. 05) were observed between the Han nationality and other minority groups. Significant correlations were observed between domestic and imported reagents, R value of CD3^+CD4^+ percentage, CD3^+CD8^+ percentage and their ratios were 0. 970,0. 953,0. 967. The u test indicated that CD3^+CD4^+ percentage had no statistically significances (P〉 0.05) while CD3^+CD8^+ percentage and their ratios had statistically significances(P〈0. 01)). Conclusions Reference values of CD3^+ CD4^+ and CD3^+CD8^+ percentage for healthy blood donors in Beijing were established initially, which would be influenced by the age ,gender, nationality, reagent and equipment. The results of domestic reagent were basically the same with those of imported reagent and significant correlations were observed between them.
