毛细管电泳法同时检测刺五加叶中的芦丁和金丝桃苷含量 被引量:6

Simultaneous Determination of Rutin and Hyperin in Acanthopanax senticosus(Rupr. et Maxim) Leaves by High-Performance Capillary Electrophoresis

摘要 目的建立简单、快速且能同时分离测定刺五加叶中芦丁、金丝桃苷含量的毛细管电泳法。方法采用毛细管电泳法,缓冲体系为0.2mol.L-1NaH2PO4,0.1mol.L-1Borax及双蒸水(10:7:3,pH8.8),熔融石英毛细管柱(25μm×45cm),分离电压为20kV,检测波长为254nm,进样时间为30s,进样高度10cm。结果芦丁、金丝桃苷在125~750,200~1200mg.L-1与峰面积线性关系良好,线性方程分别为:Y=144145ρ+423.8(r=0.9999)及Y=216836ρ-191.57(r=0.9999),样品平均回收率为99.38%及100.39%。结论该方法简单、快速、准确、重现性好,不同生长时期、不同产地的刺五加叶中两成分均存在一定的差异。 OBJECTIVE To develop a simple and rapid high-performance capillary electrophoresis (HPCE) coupled with UV detection method for the determination of rutin and hyperin in Acanthopanax senticosus ( Rupr. et Maxim ) leaves simultaneously. METHODS The buffer was composed of 0. 2 mol·L^-1 NaH2PO4 , 0. 1 mol·L^-1 Borax and double distilled water ( 10: 7: 3, pH 8. 8) solution. Capillary electrophoresis was performed using a 25 μm×45 cm fused silica capillary column. Separation voltage was 20 kV,detection wavelength was set at 254 nm , and the injection time was 30 s at a height of 10 cm. RESULTS The linear range of rutin was from 125 -750 mg·L^-1 , its linear regression equation was Y= 144 145p +423.87 (r =0. 999 9), and the average recovery was 99.38%, while the linear range of hyperin was from 200 - 1 200 mg·L^-1 , its linear regression equation was Y = 216 836p - 191.57 ( r = 0. 999 9), and the average recovery was 100. 39%. CONCLUSION This method can be used for the quality control of the Acanthopanax senticosus (Rupr. et Maxim ) leaves with good precision. The contents of rutin and hyperin are different in Acanthopanax senticosus ( Rupr. et Maxim) from different places and different collection periods.
