摘要 目的探讨缺氧条件下玉米黄质对体外培养视网膜色素上皮细胞生长的影响。方法建立人视网膜色素上皮细胞的缺氧模型(缺氧培养,体积分数为95%N2和体积分数为5%CO2),用玉米黄质进行低、中、高不同剂量(10-81、0-7、10-6mol.L-1)干预。测定对照组、缺氧模型组和玉米黄质各剂量组总活性氧(ROS)产生量、脂质过氧化物(LPO)、总超氧化物歧化酶(SOD)活性以及细胞活力。结果缺氧条件下视网膜色素上皮细胞可产生大量活性氧自由基;与缺氧组模型相比,玉米黄质各剂量组可有效抑制ROS产生量和LPO,提高总SOD活性和细胞活力(P<0.01)。结论玉米黄质可有效淬灭缺氧条件产生的活性氧,从而保护视网膜上皮色素细胞免受损伤。 Objective To investigate the effect of zeaxanthin on damage of retinal pigment epithelium cells induced by hypoxia. Methods ARPE19 cells were randomly divided into five groups: the control group,the hypoxia model group (95% N2 and 5% CO2, O2 〈 1%), the low dose (1 × 10^-8mol·L^-1 ) zeaxanthin group, the middle dose (1 × 10^-7mol·L^-1 zeaxanthin group and the high dose (1 × 10^-6mol·L^-1 ) zeaxanthin group. Cell activity, superoxide dismutase (SOD) activity, and the contents of reactive oxygen species (ROS) and lipid oxygen (LPO) were determined. Results Compared with the hypoxia model group, cell activity and SOD activity in the zeaxanthin groups were significantly increased ( P 〈 0.01 ). However, the contents of ROS and LPO were significantly decreased( P 〈 0.01 ). Conclusion Zeaxanthin can decrease the influence of active oxygen and protects retinal pigment epithelium cells against damage.
