组蛋白甲基转移酶SMYD3在不同HBV表达水平肝癌细胞中的差异表达 被引量:3

摘要 目的:探讨HBV组蛋白甲基转移酶SMYD3在肝癌细胞中的表达与HBV感染状态相关性.方法:选取HBV阴性和HBV阳性的肝癌细胞株HepG2、HepG2.2.15,利用实时逆转录聚合酶链反应(real time RT-PCR)方法检测SMYD3 mRNA表达水平,Western blot检测细胞中SMYD3蛋白表达差异.结果:SMYD3 mRNA及蛋白在HepG2.2.15的表达水平显著高于HepG2(0.92±0.12 vs 0.18±0.05,0.28±0.03 vs 0.54±0.05,均P<0.01),有统计学差异.结论:HBV可能通过上调SMYD3途径促进肝癌的恶性生物学行为. AIM: To evaluate correlation between histone methyltransferase SMYD3 expression in hepatoma and HBV infection. METHODS: SMYD3 mRNA expressions and SMYD3 protein expression levels in HBV-negative HepG2 and HBV-positive hepatoma cell line HepG2.2.15 were determined using real time PCR and Western blot, respectively. RESULTS: SMYD3 mRNA and protein levels were significantly higher in HepG2.2.15 than those in HepG2 (0.92 ± 0.12 vs 0.18 ± 0.05, 0.28 ± 0.03 vs 0.54 ± 0.05, both P 〈 0.01). CONCLUSION: HBV may promote hepatoma cell malignancy through its SMYD3 up-regulating pathways.
