摘要
目的:观察exendin-4类似物ip对链脲佐菌素(STZ)造模大鼠的血糖和ERG的影响。方法:SD大鼠36只被分成3组:正常组8只、糖尿病每日ip注射生理盐水组8只和exendin-4类似物0.05μg/g治疗组20只。检测exendin-4类似物注射期间血糖水平,糖尿病病程1mo和3mo时ERG和OPs的表现。结果:STZ造模大鼠连续exendin-4类似物注射期间平均血糖和正常组差异无统计学意义(P>0.05);而与注射用水组比较血糖明显降低(P<0.05);糖尿病大鼠ERG的b波、OPs振幅随病程进展有下降趋势,且潜伏期延长;ex-endin-4类似物治疗组的下降幅度明显小于注射用水组,统计学有差异(P<0.05)。结论:Exendin-4类似物对STZ造模大鼠有降低血糖的作用,而且可改善糖尿病大鼠ERG表现。
AIM : To observe the effect of exendin-4 analog on blood glucose in diabetic rats induced by streptozotocin (STZ) and the influence on their electroretinogram (ERG).
METHODS: Thirty-six rats were included in the experiment. The rats were divided into 3 groups: normal control (N, n=8), diabetic control (D, n=8) and E4atreated group (E4a, n = 20). For E4a group, the rats were treated with E4a S.C. (0.05ug/g BW/d). For N and D groups, the rats were treated with normal saline (NS). S.C. Blood glucose levels and body weight was measured weekly. ERGs were recorded 1 and 3 months after injection.
RESULTS: The difference of blood glucose in diabetic rats between the groups with E4a treated and the normal control was of no significance (6.68 ± 0.37 vs 6.95 ± 0.41mmol/L, P〉 0.05). The blood glucose of E4a group was much lower than that of D group. The amplitude of b-wave and oscillatory potentials (OPs) showed decrease with the progress of diabetes. The decrease in E4a treated group was less than that in untreated ones and had statistical significance ( P 〈 0.05).
CONCLUSION: E4a can normalize the blood glucose of diabetic rats. Diabetes leads to the progressive decreases in the amplitudes of b-wave and OPs. E4a can reverse them nearly to their normal condition.
