牛膝多糖对小鼠肝癌H22细胞bcl-2和Fas基因表达的影响 被引量:2

摘要 目的:研究牛膝多糖(ABPS)对肝癌H22细胞bcl-2和Fas基因mRNA表达的影响。方法:用肝癌H22细胞进行体外培养,采用MTT法检测不同浓度ABPS对H22细胞的生长抑制作用;RT-PCR检测bcl-2和Fas基因mRNA表达的变化。结果:ABPS对肝癌H22细胞具有显著生长抑制作用,并呈量效关系;bcl-2基因mRNA表达水平随着ABPS浓度的增加而下降,Fas基因mRNA表达水平随着ABPS浓度的增加而上升。结论:ABPS可抑制H22细胞增殖并诱导其凋亡,其作用机制可能与下调bcl-2基因mRNA表达、上调Fas基因mRNA表达有关。 Objective: To investigate the Effects of Achyranthes bidentata polysaccharides on Hepatocarcinoma cell line H22 and the expression of bcl-2 and Fas.Methods:Hepatocarcinoma cell line H22 were used in vitro cell culture. The inhibitory effects of ABPS on H22 cell line were assayed with MTT test. The expression of bcl-2 and Fas were detected by RT-PCR. Results: ABPS can obviously inhibit the H22 cell growth with dose dependent. The expression of bcl-2 mRNA was decreased and the expression of Fas mRNA was increased with increasing of ABPS concentration. Conclusion:ABPS may inhibit H22 cells proliferation and induce apoptosis. The possible mechanism may be through down-regulation the expression of bcl-2 and up-regulation the expression of Fas.
