摘要 目的通过对系统性红斑狼疮(SLE)患者抗热休克蛋白90抗体(HSP90-Ab)的检测,探讨其在SLE发病中的作用。方法采用Western blot法检测45例SLE患者抗HSP90抗体的表达情况,并分析其与HSP90蛋白表达间的相关性。另取15例健康血样进行正常对照。结果45例SLE患者血浆中HSP90 IgG抗体阳性表达23例(51.11%),HSP90 Ig M抗体阳性表达12例(26.67%)。并且HSP90蛋白与HSP90 IgG抗体间呈显著的正相关关系(χ2=4.15,P<0.05)。15例正常对照组中未见HSP90 IgG/Ig M抗体阳性表达。结论SLE患者体内高表达的HSP90可诱导产生抗HSP90自身抗体,其在SLE的自身免疫损伤机制中可能发挥重要作用。 Objective To investigate the role of heat shock protein 90 antibody (HSP90-Ab) in the pathogenesis of systemic lupus erythematosus (SLE) through detecting the expression of HSP90-Ab in the serum from SLE patients. Methods Western blot was used to determine IgG and IgM antibodies to HSP90 in serum of 45 SLE patients and 15 healthy controls, and to analyze the relationship between HSP90 and HSP90-Ab. Results In SLE patients, the expression rate of IgG and IgM antibodies to HSP90 was 51.11% (23 cases) and 26.67% (12 cases), respectively. No expression of IgG and IgM antibodies to HSP90 in normal serum specimens was observed. The elevated levels of HSP90 were significantly correlated with the elevated levels of IgG antibodies to HSP90 (X2 =4.15, P〈0.05). Conclusion In SLE patients, the overexpression of HSP90 protein could result in the production of HSP90-Ab, which might play a certain role in the pathogenesis of SLE.
