摘要
研究氮离子注入法诱变选育ε-聚赖氨酸高产菌株。本文从土壤筛选的菌株TS02出发,通过注入不同剂量30keV氮离子,辅以抗性平板法结合抑菌圈法,筛选出了一株ε-聚赖氨酸高产白色链霉菌(Streptomyces albulus)GC11。经多次传代实验结果表明GC11稳定性良好。摇瓶发酵培养24h,GC11抑菌圈直径为23.20mm,比TS02提高了1.32倍;5L自控发酵罐发酵培养72h,GC11ε-PL产量为4.21g/L,比TS02提高了1.30倍,达到了提高ε-聚赖氨酸产量的目的。
The screening of high ε- polylysine(ε-PL)-producing strains was studied by mutation with nitrogen ion implantation. After implantation of different dose of 30keV nitrogen ions and using inhibitive circle screening method and resistant mutant assay, a Streptomyces albulus GC11 strain with high ε-PL yield was obtained with stable genetic characteristics. After 24 h rotation-flask cultivation, the diameter of inhibition zone of GC11 strain was 23.20 mm which was 1.32-fold than that of TS02 strain; and after 72 h 5 L automatic fermentation jar cultivation, the productivity of ε-polylysine ofCJC 11 strain was 4.21 g/L which was 1.30-fold than that of TS02 sWain.
