摘要
目的比较天狼星红苦味酸染色法和MASSON三色染色法用于大鼠肾脏标本胶原纤维染色的效果。方法建立大鼠单侧输尿管梗阻肾间质纤维化模型,取梗阻侧肾脏标本,行天狼星红染色和MASSON三色染色,图像分析软件测定肾间质胶原面积。结果天狼星红染色和MASSON三色染色都能显示出胶原纤维,两种方法测定胶原面积无显著差异。天狼星红染色标本在偏光显微镜观察能分辨胶原类型。结论天狼星红苦味酸染色法比MASSON三色染色法操作步骤简单,是显示胶原的优良方法。
Objective To compare the effect of collagen fiber staining between Sirius Red F 3B(SR) in saturated carbazotic acid staining and MASSON trichrome staining on rat kidney specimen. Methods To establish renal interstitial fibrosis model of halfureteral obstruction in rat, then to get kidney specimen in obstruction side and carry out Sirius Red F 3B(SR) in saturated carbazotic acid staining and MASSON triehrome staining. Eventually, we measured the area of renal interstitial collagen by imaging analysis software. Results Both Sirius Red F 3B (SR) in saturated carbazotic acid staining and MASSON trichrome staining displayed collagen fiber. They had no significant difference in measuring the area of renal interstitial collagen. Furthermore, Sirius Red F 3B(SR) in saturated carbazotic acid staining could distinguish the collagen types through polarizing microscope. Conclusion The operation procedures of Sirius Red F 3B (SR) in saturated carbazotic acid staining are simpler than those ofMASSON trichrome staining and it is a fine method to display collagen.
