摘要
目的建立测定灵芝孢子粉中的两个有效成分灵芝三萜及灵芝多糖含量的方法,为进一步开发和应用灵芝孢子粉提供依据。方法以灵芝酸C2为标准品,通过HPLC法对灵芝孢子粉中灵芝酸C2的含量进行测定。色谱条件为:检测波长254nm;流动相为甲醇:水=55∶45,各加1%冰醋酸;柱温40℃;流速0.7mL/min。以葡萄糖为标准品,采用硫酸-蒽酮比色法,通过UV-VIS法对灵芝孢子粉中灵芝多糖的含量进行测定。结果HPLC法测得三批灵芝孢子粉中灵芝酸C2含量约为0.129‰,0.108‰和0.094‰,灵芝酸C2在7.35~147μg/mL范围内,浓度与峰面积呈良好的线性关系,r=0.9999,回收率为95.9%,95.6%和96.4%。UV-VIS法测得三批灵芝孢子粉中灵芝多糖的含量约为1.152%,1.129%和1.007%,葡萄糖在14.9~119.1μg/mL的范围内,浓度和吸光度呈线性关系,r=0.9991,回收率为95.0%,95.1%和95.2%。结论方法简便、准确,可用于灵芝孢子粉中灵芝三萜和灵芝多糖的含量测定。
Objective To establish methods for the determination of concentrations of triterpene constituents and polysaceharides in Ganoderma Lucidum Spores.Methods The HPLC condition was consisted of a RP-C18 column with a column temperature at 40℃, a mobile phase of 1% perchloric-methanol: 1% perchloric-water (55:45) with a flow rate of 0.7mL/min and detection at 254nm to determinate Ganoderma acid C2 in Ganoderrria Lucidum Spores. UV-VIS with Glucose standard was used for the determination of concentrations of polysaccharides.Resullis Tile concentrations of Ganoderma acid C2 in three batches of Ganoderma Lucidum Spores were 0.129‰, 0.108‰ and 0.094‰, and the linear range of Ganoderma acid C2 ranged from 7.35μg/mL to 147μg/mL with correlation coefficient of 0.9999. The average recoveries were 95.9%, 95.6% and 96.4%. And the concentrations of polysaccharides in three batches of Ganoderma Lucidum Spores was 1.152%, 1.129%and 1.007%, and the linear range of Glucose standard ranged from 14.9μg/mL to 119.1 μg/mL with correlation coefficient of 0.9991. The average recoveries were 95.0%, 95.1% and 95.2%. Conclusion The methods are applicable for quality control of Ganoderma Lucidum Spores.
