补肾法对肾虚排卵抑制模型大鼠内分泌激素、脏器湿重及抑制素影响的研究

Establishment and Mechanism Study of Type 1 Diabetic Animal Model of Yinyang Deficiency

目的:研究补肾法对肾虚排卵抑制模型大鼠内分泌激素、脏器湿重及抑制素的影响。方法:采用羟基脲灌胃复制肾虚排卵抑制大鼠模型,造模成功后予以罗氏促排卵汤和/或针灸补肾法治疗,治疗后观察大鼠内分泌激素、脏器湿重及抑制素的变化。结果:羟基脲造模可以使模型大鼠出现肾虚兼脾虚表现,卵巢病理切片提示排卵抑制;针刺加中药治疗后大鼠肾虚兼脾虚症状改善,内分泌激素及脏器湿重均有不同程度的增加,治疗后大鼠体重明显增加,子宫、卵巢湿重增加,差异有显著性意义(P<0.05);内分泌指标、抑制素无显著性差异(P>0.05)。结论:补肾法(包括补肾复方和/或针刺加电针)对肾虚-排卵抑制病理模型大鼠进行治疗,可以有效增加子宫及卵巢湿重,但对大鼠其它脏器湿重、内分泌及抑制素指标无明显影响。

Objective:To establish type 1 diabetic animal model of yin - yang deficiency, and to observe the changes of Cortisol,free triiodothyronine(FT3),insulin (INS),throboxane B2,and 6 - Keto -PGF1αlevels in the animal model.Methods: Sixty SD rats received intraperitoneal injection of streptozocin for the establishment of diabetic model(blood sugar over 16.7mmol/ L).After the modeling,the rats were given herbal decoction of Pericarpium Citri Reticulatae Viride,Fructus Aurantii,Dried Rhizoma Zingiberis and Cortex Phellodendri to induce yin - yang deficiency.Meanwhile, the rats received Jinkui Shenqi Bolus(JSB) for counter - evidence treatment. Results:Plasma of TXB2 and 6 - keto - PGF1αlevels as well as their ratio were lower in the diabetic model group and yin - yang deficiency model group than those in the normal group(P>0.05).In JSB group,TXB_2 level was increased(P<0.05 compared with the diabetic model group),but 6 - keto - PGF1αlevel remained unchanged (P>0.05).The cortisol level was higher in the diabetic model group,yin - yang deficiency model group and JSB group than that in the normal group(P<0.05),but was lower in JSB group than that in yin - yang deficiency model group.FT3 was lower in the diabetic model group and JSB group than that in the normal group(P<0.05). Insulin level remained unchanged in the diabetic model group,and was decreased in JSB group after treatment(P<0.01).Obvious pathological changes were found in the pancreatic islet,kidney and adrenal glands of the diabetic animal models. Conclusion:Type 1 diabetic rat models of yin - yang deficiency can be established successfully by gastric gavage of herbal decoction,which can be confirmed by the counter - evidence treatment with JSB.The mechanism of establishing the model with herbal medicine may be related with the increase of cortisol and with decrease of FT3.
