摘要
目的:观察原白头翁素衍生物(溴甲基呋喃酮)作用于人乳腺癌MCF-7细胞后Livin的表达率的变化及其意义。方法:用体外培养法培养人乳腺癌MCF-7细胞株,采用免疫细胞化学法检测Ki-67抗原和Livin蛋白表达情况。结果:原白头翁素衍生物体外对乳腺癌细胞有抗增殖作用,免疫细胞化学法分析表明原白头翁素衍生物能下调Ki-67抗原和Livin的表达。结论:原白头翁素衍生物对人乳腺癌MCF-7细胞有抑制作用,其机制可能与Livin下调相关。
Objective To study the inhibitory effect of isomycin derivant on human breast carcinoma cells in vitro and to investigate the molecular mechanism of the apoptosis.Method Human breast carcinoma cells were observed after treated with isomycin derivant.Expression of Ki-67 and Livin was determined by SABC method.Results SABC analysis showed that the expression level of Ki-67 was down-regulated by isomycin derivant;the expression of Livin was also down-regulated.Conclusion Isomycin derivant could inhibit tumor growth in human breast cells through inducing apoptosis.This action may be mediated by down-regulation of Livin.
