摘要
目的探讨在缺氧共培养条件下血管内皮细胞对绒毛外滋养细胞侵袭力的影响。方法绒毛外滋养细胞在缺氧下单独培养作为对照组,绒毛外滋养细胞与血管内皮细胞用Transwell小室建立缺氧共培养作为观察组。应用荧光定量PCR、Western blot方法分别检测绒毛外滋养细胞基质金属蛋白酶9(MMP9)/基质蛋白酶抑制剂1(TIMP1)mR-NA及蛋白的表达;应用Transwell侵袭模型检测缺氧共培养下绒毛外滋养细胞侵袭力的变化。结果与对照组比较,观察组的绒毛外滋养细胞MMP9mRNA及蛋白的表达均明显降低(均P<0.01),TIMP1mRNA及蛋白的表达无明显变化(均P>0.05);观察组的绒毛外滋养细胞侵袭力较对照组明显降低(P<0.01)。结论缺氧共培养条件下,血管内皮细胞可下调绒毛外滋养细胞MMP9基因的表达并降低绒毛外滋养细胞侵袭力。
Objective To investigate the invasive effect of extravillous trophoblast cells by hypoxic co-cultured vascular endothelial cells.Methods The control group was extravillous trophoblast cells that were lonely cultured under hypoxic condition.The experimental group was extravillous trophoblast cells and vascular endothelial cells that were co-cultured under hypoxic condition in Transwell chamber.Fluorescence quantitative real-time PCR and Western blot were used to determine the mRNA and protein expression of MMP9 and TIMP1 in extravillous trophoblast cells.The invasion of extravillous trophoblast cells was assessed by Transwell chamber assay.Results The mRNA and protein of MMP9 were more down-regulated in the experimental group than in the control group(both P〈0.01),and the mRNA and protein of TIMP1 had no significant changes(P〉0.05).The invasion of extravillous trophoblast cells in the experimental group was more decreased than in the control group(P〈0.01).Conclusion Vascular endothelial cells can down-regulate the MMP9 gene expression of extravillous trophoblast cells and reduce the invasion of extravillous trophoblast cells under hypoxic co-cultured conditions.
