摘要
以米根霉(Rhizopus oryzae)CS-323为出发菌株,经过紫外线(UV)诱变后,利用溴钾酚绿平板变色圈法及高锰酸钾-溴化钾平板透明圈筛选出18株变异株。基于上述方法,设计琼脂块溴钾酚绿变色圈法及琼脂块高锰酸钾-溴化钾透明圈法,对18株变异株进行进一步筛选,经摇瓶初筛验证,琼脂块透明圈法能准确且迅速的判断菌株产酸大小,并对平板定量初筛检出高产株有较大的意义。
The original strain Rhizopus oryzae CS-323 was mutated by UV,and 18 mutants were selected by bromocresol green agar plate and potassium permanganate-potassium bromide agar plate.Base on above,a agar block filtration method was designed and used on those mutants.Verified by shaking flask fermentation,the agar block method can quickly and exactly select the high yield mutants,and have notable significance of quantitative plate filtration.
