摘要
目的:观察平糖方药物血清对高浓度软脂酸刺激下的INS-1β细胞胰岛素分泌的影响。方法:以大鼠胰岛β细胞株INS-1细胞为模型,以平糖方药物血清作为刺激因素,通过MTT方法,测定INS-1细胞的生长和增殖能力;在0.5mmol/L PA的条件下,分别观察平糖方药物血清对INS-1细胞胰岛素分泌和ATP合成的影响,胰岛素分泌采用RIA方法检测,ATP采用荧光素酶方法检测。结果:平糖方药物血清对INS-1β细胞的生长增殖有一定的抑制趋势,其中高剂量组与细胞对照组相比出现了明显的降低。PA作用于细胞24h可以导致胰岛素分泌增加、ATP合成升高,平糖方药物血清可使PA促进的胰岛素分泌降低、ATP合成降低;而作用于细胞72h可以导致胰岛素分泌减少、ATP合成降低,平糖方药物血清对PA引起的ATP降低没有明显的改善作用。结论:平糖方药物血清急性抑制PA促进胰岛素分泌和ATP合成,还可能通过抑制ATP合成而急性抑制PA促进胰岛素分泌。
Objective:To investigate the effect of fomula Pingtang drug-containing serum on insulin secretion of INS-1 β cell chronically and acutely exposured to high concentrations palmitic acid.Methods:INS-1 β cell were incubated for 24h and 72h in a medium containing fomula Pingtang drug-containing serum and 0.5 mmol /L palmitic acid.Cell proliferation capacity were measured by MTT.Insulin contents in the medium were measured by radioimmunoassay.ATP contents of cells were measured by chemiluminescence method.Result:Fomula Pingtang drug-containing serum of the high and middle dose group inhibited INS-1 cell growth and proliferation.Fomula Pingtang drug-containing serum lowered insulin secretion of β cell acutely exposured to high concentrations of PA and reduced the amount of ATP synthesized.Conclusion: Fomula Pingtang Drug-containing Serum acutely inhibited insulin secretion and ATP contents from high concentrations of PA stimulated INS-1β cells.
