摘要
目的观察玉屏风多糖(YPF-P)对大鼠肝纤维化的保护作用。方法采用CCl4诱导大鼠肝纤维化模型。于12周末采血,测血清中丙氨酸氨基转氨酶(ALT)、天冬氨酸氨基转移酶(AST)含量;取部分肝组织做病理组织学和羟脯氨酸(Hyp)、丙二醛(MDA)、超氧化物歧化酶(SOD)含量测定;免疫组化法检测核转录因子-κB(NF-κB)和转移生长因子-β1(TGF-β1)蛋白表达;逆转录聚合酶链反应(RT-PCR)法检测TGF-β1mRNA的表达。结果 YPF-P(120、60mg/kg)组能明显改善肝脏组织结构,减轻肝纤维化增生程度,同时显著降低肝纤维化大鼠血清中ALT、AST活性和肝组织中Hyp、MDA含量,升高SOD含量。此外,YPF-P(120、60mg/kg)组也能明显抑制肝组织中NF-κB、TGF-β1蛋白和TGF-β1mRNA的表达。结论 YPF-P抑制纤维化大鼠肝组织NF-κB和TGF-β1的表达可能是其抗肝纤维化主要作用机制之一。
Objective To observe the effects of polysaccharides of Yu-Ping-Feng Powder(YPF-P) on CCl4-induced hepatic fibrosis in rats. Methods Liver fibrosis was induced by CCl4 twice a week in rats for 12 weeks. YPF-P (120,60,30 mg/kg) was treated daily via gastrogavage. The levels of ALT,AST in serum and Hyp,MDA,SOD contents in liver tissue were detected using the kits. Liver tissue samples were stained with hematoxylin-eosin ( HE) . TGF-β1 and NF-κB protein expression were determined by immuninochemistry. TGF-β1 mRNA expression was detected by RT-PCR. Results Compared with model group,YPF-P ( 120,60 mg/kg) treatment significantly reduced serum ALT,AST and liver MDA、Hyp content,and increased SOD in liver tissue. YPF-P ( 120,60 mg/ kg) effectively reduced the protein expression of NF-κB and TGF-β1 as well as TGF-β1 mRNA. Conclusion YPFP can inhibit expression of TGF-β1 and NF-κB,which is probably one of the anti-hepatic fibrosis mechanisms of YPF-P.
