基于快速高分辨液相色谱串联质谱技术的代谢组学尿液分析方法的建立 被引量:21

摘要 采用快速高分辨液相色谱(RRLC)分离系统与QTRAP型及QTOF型MS/MS仪联用技术,通过考察尿液样本前处理方法,优化液相色谱条件和质谱检测参数,建立了用于尿液中代谢物分析的RRLC-MS方法。采用本方法对尿液浓度下的20种代表性代谢物进行了检测,考察了方法的灵敏度和精密度,证明本方法适用于尿液代谢组学的研究。对穿插在检测序列中的生物质控样本的监测结果表明,本方法的可重复性及获得的数据的可靠性良好。本方法已成功地应用于乳腺癌和宫颈癌的尿液代谢组学研究以及可能生物标志物的检测。 A rapid resolution liquid chromatography/tandem mass spectrometric(RRLC-MS/MS) method for urinary metabolites analysis has been implemented on a quadrupole-linear trap(QTRAP) and quadrupole-time of flight(QTOF) type instrument in both positive and negative ion modes.The pretreatment methods for urine samples and the analytical condition of liquid chromatography and mass spectrometry analysis have been studied.The sensitivity and precision of the established LC-MS method have been examined by using twenty representative metabolites at urine concentration.The results indicated that the method can be used for metabono-mics analysis.Moreover,by carefully monitoring of the quality control samples randomly inserted amongst the real sample queue,it demons-trate that the reproducibility of this method is sufficient to ensure data quality in metabono-mics applications.The method has been successfully employed in the urinary metabonomics study on breast cancer and uterine cervix cancer.Several biomarkers have been detected and identified.
