摘要
目的:比较Folin-酚(Lowry)法、双辛可宁酸(BCA)法、考马斯亮蓝(Bradford)法测定蛋白含量的的准确性及适用性。方法:采用上述3种蛋白浓度测定方法测定标准小牛血清蛋白(BSA)含量,考察其灵敏度、准确度、精密度、线性范围及影响因素,并用于大鼠肝微粒体蛋白含量的测定。结果:3种方法线性及相关度都较好,线性范围以BCA法最宽,Bradford法次之,但Lowry法、BCA法精密度及准确度都优于Bradford法,抗离子干扰能力也明显优于Bradford法。结论:BCA法测定肝微粒体蛋白浓度较准确、稳定,但Lowry法操作简单,原料经济,准确性及稳定性也较好,更适合于实验室研究使用。
Objective: To compare the accuracy and applicability of protein content determination by Lowry method, BCA method and Bradford method. Method: The concentration of bovine serum albumin (BSA) protein standards was determined by the three methods to examine their sensitivity, accuracy, precision, linear range and impact factors. The methods were also applied in rat liver microsomal protein content determination. Result: Three methods all had good correlation linearity, and BCA method had the largest linear range. The precision, accuracy and ion interference resistance of Lowry method and BCA method were both better than the Bradford method. Conclusion: BCA method to determine the concentration of rat liver mierosomes is more accurate and stable, but Lowry method is easier to operate with cheaper raw materials ,which is more suitable for laboratory research.
