摘要
[目的]探讨人参皂甙Rh2对肝癌Bel-7402细胞侵袭和迁移的影响及其机制.[方法]采用MTT法检测人参皂甙Rh2对肝癌Bel-7402细胞活力的影响;Transwell小室测定侵袭和迁移能力;应用Western blot方法检测细胞基质金属蛋白酶(MMP)-2和MMP-9蛋白水平的变化.[结果]给予人参皂甙Rh2处理后肝癌Bel-7402细胞生长受到抑制,细胞侵袭和迁移能力明显降低;肝癌Bel-7402细胞MMP-2和MMP-9蛋白的表达水平均明显减弱.[结论]人参皂甙Rh2可抑制肝癌Bel-7402细胞的侵袭和转移能力,其机制可能与降低MMP-2和MMP-9蛋白表达水平有关.
OBJECTIVE To explore the effects of gensenoside Rh2 and its mechanism on the invasion and migration of the human hepatic carcinoma Bel-7402 cells.METHODSThe effects of gensenoside Rh2 on cell viability was tested by MTT,and the cell invasion and migration were determined by transwell chamber model,and the expression of matrix metalloproteinases-2(MMP-2) and MMP-9 were detected by Western blot.RESULTSAfter being treated with gensenoside Rh2,the proliferation of Bel-7402 cells was inhibited,and the invasion and migration of Bel-7402 cells were also inhibited.The expression levels of MMP-2 and MMP-9 were decreased.CONCLUSIONThe gensenoside Rh2 could inhibite the invasion and migration of the human hepatic carcinoma Bel-7402 cells,and its mechanism is probably correlated with decrease of MMP-2 and MMP-9 expression.
