摘要
为了探讨毒害艾美耳球虫(E.necatrix)感染雏鸡后,其免疫器官白细胞介素(IL-2)和T、B淋巴细胞增殖能力的变化,本研究应用组织匀浆涂片和酸性α-醋酸萘酯酶(ANAE)染色及细胞培养技术和四甲基偶氮唑盐(MTT)测定法对E.necatrix感染雏鸡免疫器官的T淋巴细胞百分数、IL-2诱生活性、T和B淋巴细胞对ConA或PMA的增殖能力动态变化进行较全面系统的研究。结果发现,E.necatrix感染雏鸡后,其胸腺和脾脏T淋巴细胞百分数分别于感染后7d~21d和7d~24d明显高于对照雏鸡;IL-2诱生活性分别于16d~18d和18d~21d较对照雏鸡显著升高;T淋巴细胞对ConA的增殖能力分别在14d~16d和10d~18d明显增强。法氏囊和脾脏B淋巴细胞对PMA的增殖能力分别于14d~24d和14d~21d显著高于对照雏鸡。表明E.necatrix感染雏鸡免疫器官的IL-2调节及细胞免疫和体液免疫功能均明显提高。
Dynamic changes of percentage of T lymphocytes, IL-2 induced activity and proliferation of T and B lymphocytes to concanavalin A (ConA) or phorble myristate acetate (PMA) in immune organs of 14-day-old chicks infected with Eimeria necatrix were studied using acid nonspecific alpha-naphthyl acetate esterase (ANAE), cellular culture technique and MTT assay. These results showed that the IL-2 induced activity in thymus and spleen of chicks infected with E.necatrix was significantly increased compared with that of controls at 16 to 18 day and 18 to 21 day post infection (PI), respectively. The proliferation of T lymphocytes to ConA in thymus and spleen of chicks infected with E. necatrix was significantly elevated compared with control chickens at 14 to 16 day and 10 to 18 day PI, respectively. The proliferation of B lymphocytes to PMA in spleen and bursa of Fabricius of chicks infected with E. necatrix was significantly higher than that of control at 14 to 24 day and 14 to 21 day PI, respectively. These results showed that IL-2 immune regulation and function of cellular and humoral immunity in immune organs of chicks infected with E. necatrix had been significantly increased.
