Simultaneous Determination of Ferulic Acid,5-Methoxypsoralen,Imperatorin and Isoimperatorin in Duliang Dropping Pills by High Performance Liquid Chromatography

目的:建立一种同时测定都梁滴丸中的阿魏酸、5-甲氧基补骨脂素、欧前胡素、异欧前胡素含量的方法。方法:都梁滴丸以75%甲醇超声提取30 min后取上清液,采用Agilent Zorbax SB C18(4.6 mm×150 mm,5μm)色谱柱,以甲醇(A)-0.1%磷酸水溶液(B)为流动相进行梯度洗脱,流速1.0 mL.min-1;检测波长分别为254,320,330 nm。结果:4种成分的方法最低定量限为3～15 mg.kg-1,线性范围0.5～50 mg.L-1,相关系数(r2)≥0.999 5。高、中、低3个添加水平的平均加样回收率(n=5)为96.6%～102.5%,RSD 0.16%～2.07%。结论:该方法简便、快速适用于都梁滴丸中阿魏酸、5-甲氧基补骨脂素、欧前胡素和异欧前胡素的含量测定。

Objective： To develop a high performance liquid chromatographic（HPLC） method for simultaneous determination of ferulic acid,5-methoxypsoralen,imperatorin,isoimperatorin in Duliang dropping pills.Method： Duliang dropping pills were extracted supersonically with 75% methanol for 30 min.The supernatant was separated on Agilent Zorbax SB C18（4.6 mm×150 mm,5 μm） by gradient program with methanol（A）-water with 0.1% phosphoric acid（B） as the mobile phase at a flow rate of 1.0 mL·min-1,and determined by diode array detector at 254,320,330 nm respectively.Result： The limits of quantification of four compounds were in the range of 3-15 mg·kg-1,this method showed a good linearity between 0.5-50 mg·L-1 and the correlation coefficient（R2） was more than 0.999 0.The average recoveries were from 96.6% to 102.5% with relative standard deviation from 0.16% to 2.07%.Conclusion： This method is simple and rapid and suitable for determination of the four compounds in Duliang dropping pills.
