摘要 目的:建立氧化石墨(GO)纳米粒中10-羟基喜树碱(HCPT)含量测定的高效液相色谱方法。方法:色谱柱:Diamonsil^(TM)ODS-C_(18)(200 mm×4.6 mm,5μm),流动相:乙腈-水(25:75),流速:1.0 ml·min^(-1),检测波长:266 nm。结果:在本试验条件下制剂中辅料对HCPT含量的测定无干扰,HCPT的质量浓度在1~100μg·ml^(-1)内线性关系良好(r=0.999 9),平均回收率为99.54%。GO纳米粒中HCPT含量可达到(0.157±0.009)mg·mg^(-1)。结论:含量测定方法准确可靠、简单易行,可用于GO纳米粒中HCPT的含量测定。 Objective: To establish an HPLC method for the determination of 10-hydroxycamptothecin (HCPT)-loaded graphite oxide nanoparticles. Method: DiamonsilTM ODS-Cls column (200 mm ×4. 6 ram,5 μm) was used with aeetonitrile-water (25: 75) as the mobile phase at the flow rate of 1.0 ml·min^-1. HCPT was detected at 266 nm. Result: The linear range of HCPT was 1-100μg· ml^-1 (r = 0. 999 9 ). The average recovery of HCPT was 99.54%. The amount of HCPT loaded in graphite oxide nanoparticles was as high as (0. 157 ±0. 009) mg·mg^-1. Conclusion: The method is accurate and simple for the determination of HCPT in graphite oxide nanopanoarticles.
