摘要
目的研究全反式维甲酸(ATRA)在体外对人类急性早幼粒细胞白血病(APL)细胞株NB4和HL-60细胞增殖分化和程序化死亡的作用。方法细胞形态学、流式细胞仪、核酸琼脂糖电泳等多参数进行研究。结果ATRA有诱导以上两种细胞分化、抑制细胞增殖的作用;维甲酸有诱导HL-60细胞在分化成熟后迅速产生程序化死亡的作用。结哈PML/RARα(早幼粒白血病/维甲酸α受体)融合蛋白的存在阻碍了NB4细胞在分化成熟后迅速发生程序化死亡。对阐明线甲酸的作用机理指导临床制订合理治疗方案有一定价值。
Objective Cell differentiation, proliferation and apoptosis had been inves-tigated in human promyelocytic leukemia cell line NB4 and HL- 60 cells treated with alltrans retinoid acid. Methods These processes were monitored by cell morphology, flowcytometry and DNA gel electrophoresis. Res'Its The results showed (1) All trans retinoicacids could induce cell diferentiation and inhibit cell proliferation; (2) When exposed to alltrans retinoic acids, HL- 60 cells could be induced to apoptosis right after maturation. Incontrast, NB' cells with PML/RARa fusion protein had fewer apoptosic cells after mat-uration than HL - 60. ConcIusion These data may be helpful in explaining the mechanismof retinoids effect on acute promyelocyte leukemic cells.
