摘要
目的观察人参皂甙Rh2对人乳腺癌MCF7/Adr细胞侵袭和迁移的作用及其机制。方法用MTT比色法,检测人参皂甙Rh2(Rh2)对MCF7/Adr细胞的活力;Transwell小室测定其侵袭力和迁移力;Western blot法检测细胞基质金属蛋白酶2(MMP2)、MMP9和核转录因子KappaB(NF-KB)蛋白水平的变化。结果用人参皂甙Rh2处理后,MCF7/Adr细胞生长受到抑制,且作用呈时效-量效依赖关系;同时细胞侵袭和迁移能力明显降低;MMP2、MMP9和NF-kB蛋白的表达均明显减弱。结论人参皂甙Rh2能够减弱MCF7/Adr细胞侵袭和转移,其机制可能与降低MMP2、MMP9和NF-kB蛋白表达有关。
Objective To explore the effect of Gensenoside Rh2 and its mechanism on the invasion and migration of human breast cancer MCF7ArR cells.Methods The effect of Gensenoside Rh2 on cell viability was tested by MTT.The cell invasion and migration were determined by transwell chamber modcl.The expression of matrix metalloproteinases 2(MMP-2),MMP9,and nuclear factor-kappa B(NF-kB) were detected by Western blot.Results After being treated with Gensenoside Rh2,the proliferation of MCF7/AdrR cells was inhibited in a time and dose-depending manner.The invasion and metastasis of MCF7/AdrR cells were also inhibited.The expression level of MMP-2,MMP9,and NF-kB were decreased.Conclusion Gensenoside Rh2 could affect the invasion and metastasis of the human breast cancer cells Caski.It is probably one of its molecular mechanisms to inhibit the expression of MMP-2,MMP9,and NF-kB.
