摘要
目的观察高迁移率族蛋白B1(HMGB1)在慢性阻塞性肺疾病(COPD)模型大鼠及断烟后大鼠肺部的表达。方法雄性SD大鼠随机分为正常对照组、COPD组及断烟组,每组10只。采用烟熏法建立COPD大鼠模型,HE染色观察支气管肺组织病理形态学改变,免疫组化法检测HMGB1的表达。结果 COPD组和断烟组肺平均内衬间隔显著高于正常对照组(P<0.05),而COPD组和断烟组比较差异无统计学意义;免疫组化染色可见HMGB1主要分布于支气管及肺泡上皮细胞,3组间平均积分光密度(AIOD)比较差异均有统计学意义(P<0.05)。结论单纯被动吸烟可复制COPD模型,HMGB1可能与COPD的发病有关。
Objective To evaluate the expression of high mobility group box1(HMGB1) in cigarette smoking induced rat model of COPD and after smoking cessation in rat.Methods Thirty male SD rats were randomly divided into control group,COPD model group and smoking cessation group,10 rats in each group.Pathologicol morphology change of bronchial lung tissue was observed by HE staining.The expression of HMGB1 in the lung were detected by immuno-histochemcal method.Results The mean linear intervals in COPD group and smoking cessation group were higher than those in the control group(P 〈0.05);but it was no statisticall different between COPD group and smoking cessation group.HMGB1 was found in the bronchial and alveolar epithelial cells,the AIOD in COPD group way higher than that in the control groupand smoking cessation group;and the expression in smoking cessation group was higher than that in the control group(P 〈0.05).Conclusion Cigarette smoke can induce rat model of COPD,HMGB1 may play a role in the pathogenesis of COPD.
