新型雌激素受体GPR30/GPER1及其在乳腺癌中的作用 被引量:3

摘要 G蛋白偶联受体30(G protein-coupled receptor 30,GPR30)作为一种新型雌激素受体被发现,它与经典雌激素受体没有同源性,作用机制和效应也均有不同,主要通过表皮生长因子受体转活后的蛋白激酶途径和第二信使cAMP、Ca2+介导快速非基因效应,继而调节转录因子的转录活性。在ER(-)乳癌细胞中,它促使肝素结合表皮生长因子前体pro-HB-EGF转变为HB-EGF并释放,反式激活表皮生长因子受体。它不但构建了慢速基因效应和快速非基因效应的交互通话,还沟通了雌激素和表皮生长因子,使雌激素具有表皮生长因子类似的功能,它介导了雌激素靶器官癌症细胞的发生发展,是新型的肿瘤标志物,同时促使乳癌细胞产生耐药性。本文就GPR30/GPER1的研究进展进行综述并论述它在乳腺癌中的作用,对其在乳腺癌研究和治疗中的前景进行展望。
