摘要 目的采用快速液相-高分辨四极杆飞行时间串联质谱对金银花化学成分进行分离鉴别。方法采用WatersACQUITY UPLC BEH C18色谱柱(2.1mm×50mm,1.7μm),乙腈(含0.1%甲酸)和0.1%甲酸溶液梯度洗脱,流速0.4mL.min-1,进样量为5μL,检测器为高分辨四极杆飞行时间质谱,ESI离子源,质量数扫描范围m/z100~1 500。结果根据正、负离子模式下获得的各色谱峰的质谱数据,共检测到33个化学成分,其中6个咖啡酰奎尼酸类、7个黄酮类和7个环烯醚苷类成分通过与文献对照或根据质谱裂解规律得到进一步确认。结论此方法灵敏准确,有利于化合物结构鉴别,可作为中药金银花的咖啡酰奎尼酸类、黄酮类和环烯醚苷类成分的鉴定方法。 Objective To separate and identify the chemical constituents of Lonicera japonica Thunb.by RRLC-Q-TOF/MS.Methods Waters ACQUITY UPLC BEH C18 column(2.1 mm×50 mm,1.7 μm) was used with acetonitrile(containing 0.1% formic acid)-0.1% formic acid solution as the mobile phase in gradient elution.The flow rate was 0.4 mL·min-1,the injection volumn was 5 μL,and the detector was high-resolution quadrupole time of flight mass spectrometry with an ESI ion source.Scanning mass range was m/z 100-1 500.Results According to mass spectrometry in positive and negative ion mode,33 compounds were detected,including 6 caffeoylquinic acids,7 flavonoids,and 7 iridoid glycosides,which were characterized based on their fragmentation patterns or comparison with previous references.Conclusion This method is sensitive,accurate and beneficial for compounds identification,which can be used as the identification method for caffeoylquinic acids,flavonoids,and iridoid glycosides of Lonicera japonica Thunb..
