摘要
目的:研究消癌解毒方对肝癌细胞株H22凋亡及Survivin表达的影响,探讨其可能的抗癌机制。方法:建立小鼠H22移植瘤模型,分对照组和XJR低、中、高剂量(10、30、90g/kg)组及顺铂(0.001g/kg)组进行治疗,流式细胞术检测各组移植瘤细胞周期及凋亡率,Western blot法检测H22移植瘤细胞Survivin蛋白表达。结果:消癌解毒方各剂量组和顺铂阳性对照组的小鼠平均凋亡率均高于模型组小鼠平均凋亡率。二者比较,差异均具有统计学意义(P<0.01,P<0.05)。FCM细胞周期显示,与模型组比较,消癌解毒方中剂量组和顺铂阳性对照组细胞G0/G1期比例相对增加,P<0.05,差别具有统计学意义,S期及G2从期比例相对下降,细胞周期于G0/G1期发生阻滞,P<0.05,差别具有统计学意义。Western blot法检测各组Survivin蛋白表达结果与模型组相比,各组Survivin蛋白表达均有下降趋势,其中DDP阳性对照组Survivin蛋白表达降低最明显,消癌解毒方10mg/kg低剂组、90mg/kg高剂组和30mg/kg中剂组Survivin蛋白表达也依次降低。结论:消癌解毒方能促进肿瘤细胞凋亡,抑制Survivin蛋白表达,是其抗癌作用机制之一。
Objective:To investigate the effect of Xiaoai Jiedu Recipe(XJR)on apoptosis and Survivin expression in hepatocarcinoma cell line of H22,and to study the possible anti-tumor mechanisms.Methods:Establishing the mice transplanted H22 hepatocarcinoma tumor model,control group,XJR sub-group of low,medium and high dose(10,30,90mg/kg)group and cisplatin(0.001mg/kg)treatment group were devided,we used flow cytometry technique to detect the tumor cell cycle and apoptosis in each group,Western blot assay tested the expression of Survivin protein of transplanted hepatocarcinoma H22 tumor cells.Results:The average apoptosis rate of the mice in each dose group of Xiaoai Jiedu Recipe(XJR)and cisplatin positive control group was higher than the model group mice.Comparing the two groups,the differences were statistically significant(P0.01,P0.05).FCM cell cycle showed that compared with model group,the proportion of cells in G0/G1 period of XJR medium dose group and cisplatin positive control group increased relatively,P0.05,the difference was statistically significant.By Western blot analysis,compared with model group,the expression of Survivin protein in each group were decreased,and DDP positive control group decreased obviously,followed by XJP sub-group of low,high and medium dose(30,10,90mg/kg)group,respectively.Conclusion:Xiaoai Jiedu Recipe(XJR)can promote tumor cell apoptosis and inhibit expression of Survivin protein,which is one of the mechanisms of its anti-tumor effects.
