二至丸、地黄煎影响小鼠免疫功能的比较研究 被引量:18

Comparison on Effect of Erzhi Pills and Dihuang Decoction on the Immune Function in Mice

摘要 目的:探讨二至丸、地黄煎的免疫调节作用。方法:采用KM雌性小白鼠随机分为6组:正常对照组、模型组、香菇多糖(10 mg·kg-1)组、二至丸合地黄煎组(18.2 g·kg-1)、二至丸组(7.8 g·kg-1)和地黄煎(10.4 g·kg-1)组。除了正常对照组外,其余各组均给小鼠ip环磷酰胺建立免疫低下模型,各组分别按设计剂量连续ig给药1周。检测小鼠胸腺指数、脾指数、血清白细胞介素1-α(IL-1α)水平、脾细胞的细胞周期及脾细胞凋亡率。结果:模型组小鼠胸腺指数(1.84±0.46)mg·g-1、脾指数(2.14±0.61)mg·g-1、IL-1α水平(135.21±28.69)ng.L-1均显著性降低,脾细胞增殖周期抑制[S期(2.37±3.08)%],脾细胞凋亡率高达52.86%;而二至丸合地黄煎组、二至丸组、地黄煎组及香菇多糖组均可不同程度地升高胸腺指数(二至丸合地黄煎组:(2.41±0.66)mg·g-1,二至丸组:(2.57±0.62)mg·g-1,地黄煎组:(2.47±0.63)mg·g-1、脾指数(二至丸合地黄煎组(5.17±1.58)mg·g-1,二至丸组:(4.40±0.87)mg·g-1,地黄煎组:(5.03±1.21)mg·g-1、IL-1α水平(二至丸合地黄煎组(180.72±38.94)mg·g-1,二至丸组(179.51±43.46)mg·g-1,地黄煎组(172.93±40.91)ng.L-1,可不同程度地拮抗环磷酰胺引起的脾细胞增殖抑制,二至丸合地黄煎组(15.17±2.79)%,二至丸组(9.55±3.73)%,地黄煎组(10.86±4.29)%及凋亡(二至丸合地黄煎组(5.26±7.14)%,二至丸组(8.34±3.68)%,地黄煎组(6.79±4.31)%。结论:二至丸方、地黄煎方及二至丸合地黄煎组方均可提高免疫低下实验小鼠的免疫功能,均具有抗凋亡作用,其中二至丸、地黄煎两方合用效果较单方更好。 Objective:To investigate the immunoregulation function of Erzhi pills and Dihuang decoction.Method: Female mice were adopted and divided into 6 groups: control group,model group,lentinan group(10 mg·kg-1),group of Erzhi pills and Dihuang decoction(18.2 g·kg-1),Erzhi pill group(7.8 g·kg-1) and Dihuang decoction group(10.4 g·kg-1).Except for control group,mice in other groups were given cyclophosphamidum(CY) by intraperitoneal injection to establish immunosuppression model.Drugs were given intragastrically for one week in each group continuously according to respectively-designed doses.The thymus index,spleen index,level of serum interlukin-1α(IL-1α),cellular cycle and apoptosis of spleen cells of mice were detected.Result: In model group,the thymus index(1.84±0.46)mg·g-1,spleen index(2.14±0.61)mg·g-1 and IL-1α level(135.21±28.69)ng·L-1 markedly decreased,proliferation of spleen cells were inhibited(2.37±3.08)% and their apoptosis rate was up to 52.86%,whereas the group of Erzhi pills and Dihuang decoction,Erzhi pill group,Dihuang decoction group and lentinan group could all increase the thymus index(group of Erzhi pills and Dihuang decoction(2.41±0.66) mg·g-1,Erzhi Pill group(2.57±0.62)mg·g-1,Dihuang decoction group(2.47±0.63) mg·g-1,spleen index(group of Erzhi pills and Dihuang decoction(5.17±1.58),Erzhi Pill group:(4.40±0.87),Dihuang decoction group(5.03±1.21) mg·g-1 and IL-1α level(group of Erzhi pills and Dihuang decoction(180.72±38.94) ng·L-1,Erzhi pill group(179.51±43.46)ng·L-1,Dihuang decoction group(172.93±40.91) ng·L-1 and resist the proliferation(group of Erzhi pills and Dihuang decoction(15.17±2.79)%,Erzhi pill group(9.55±3.73)%,Dihuang decoction group(10.86±4.29)% and apoptosis of spleen cells(group of Erzhi pills and Dihuang decoction(5.26±7.14)%,Erzhi Pill group(8.34±3.68)%,Dihuang decoction group(6.79±4.31)%caused by CY to varying degrees.Conclusion: Erzhi pills,Dihuang decoction and group of Erzhi pills and Dihuang decoction can enhance the immune function in immunosuppressed experimental animals and have the function of anti-apoptosis,in which the effect of Erzhi pills combined with Dihuang decoction is better than that of single one.
