摘要
通过甲基磺酸乙酯(EMS)诱变晚粳稻品种秀水09,获得了1个稳定遗传的全生育期黄绿叶突变体Ygl,遗传分析表明该突变性状由1对隐性核基因控制。利用突变体与籼稻品种珍汕97杂交,构建F2群体对突变基因进行定位,发现目的基因与第11号染色体上的SSR标记RM2459连锁,在该标记附近发展了14对InDel标记,将突变基因定位在约82kb区间内。通过在线最新水稻注释系统的基因功能预测分析,寻找候选基因,经测序发现突变体发生单碱基突变。
A stably inherited yellow-green leaf mutant was isolated by EMS mutagenesis for a Japonica rice variety Xiushui 09 and it exhibited yellow green leaf during the whole growth stage. Genetic analysis indicated that the phenotype was controlled by a single recessive nuclear gene and designated tentatively as ygl( 1 ). To map the mutant gene, an F2 population was constructed by crossing mutant with Indica rice variety Zhenshan 97. The ygl( 1 ) was linked to SSR marker RM2459 on the chromosome 11. Then, 14 InDel markers were developed around the marker, ygl( 1 ) gene was finaly mapped to an 82kb region. Candidate gene was predicted and analyzed by Rice Genome Annotation (http ://rice. plantbiology, msu. Edu/overview. shtml) , and sequencing of the candidate gene from the Ygl mutant and the wild type variety by online system ( http ://rice. rice. plantbiology, msu. Edu/overview. shtml) showed that a single nucleotide mutation occurred in the mutant.
