摘要
目的研究基质金属蛋白酶-2组织抑制剂(tissue inhibitor of metalloproteinases-2,TIMP-2)基因多态性与蛋白质表达的关系。方法采用聚合酶链反应-限制性酶切片段长度多态性(PCR-RFLP)方法检测29份无病变肺组织TIMP-2基因+853位点单核苷酸多态性(single nucleotide polymorphisms,SNPs),免疫组化法检测肺组织TIMP-2蛋白的表达,分析TIMP-2基因+853位点多态性与蛋白质表达的关系。结果 TIMP-2基因+853位点基因型频率为G/G型51.7%、G/A 31.0%、A/A 17.3%,TIMP-2蛋白表达半定量和定量检测各型间无差异性(P>0.05)。结论 TIMP-2基因+853位点基因多态性的改变不影响TIMP-2蛋白的表达。
Objective To study the relationship between TIMP-2 gene polymorphism and protein expression of TIMP-2. Methods Methods The single nucleotide polymorphism of position + 853 of the TIMP-2 gene was revealed in 29 pulmonary tissues without pathological changes by polymerase chain reaction-restriction fragment length polymorphism. Immunohistochemical method was used for the detection of protein expression of TIMP-2 in pulmonary tissues. Association between the single nucleotide polymorphism of TIMP-2 gene at posi- tion + 853 and protein expression of TIMP-2 was analyzed. Results At position + 853, the genotype frequencies were 51.7% for G/G,31% for G/A,and 17.3% for A/A. Protein expression of TIMP-2 was not significantly different in three genotypes by semiquanfitative detection and quanfitate detection ( P 〉 0. 05 ). Conclusion TIMP-2 gene polymorphism at position + 853 does not change protein expression of TIMP-2.
