摘要
目的 研究西比灵逆转抗癫痫药(AEDs)诱导的星形胶质细胞多药耐受基因(MDR1)表达的规律。方法 将苯巴比妥钠(PB)和丙戊酸钠(VPA)分别加入培养的星形胶质细胞内,30天后用免疫细胞化学法检测MDR1的表达产物P-糖蛋白(Pgp),继之将终浓度为5μg/ml的西比灵分别加入培养的正常细胞(亲本细胞)及MDR1表达增强的细胞(耐药细胞)内,用四甲基偶氮唑盐(MTT)法计算逆转倍数和相对逆转效率,了解西比灵逆转MDR1表达的作用。结果 发现西比灵对耐PB或VPA的星形胶质细胞的逆转数分别为2.09、2.51,相对逆转效率分别为76.25%、67.80%。结论 西比灵对AEDs诱导的MDR1的表达有部分逆转作用。
Objective:To study the Sibelium(Flunarizine) reverses antiepileptic drugs(AEDs)- induced overexpression of multidrug resistance gene (MDR1). Methods: Phenobarbital and Valproic acid were added into astrocytes and cultured permanently for 30 days. P-glycoprotein (Pgp); a product of MDR1, was detected with immunocyto chemistry.Then,5μg/ml Sibelium was added to resistance cells(Pgp positive cells) and nonresistance cells(umstimulated by AEDs),MTT assay was used to measure reverse multiple and relative reverse efficiency rate.Results:Our study showed reverse multiples were 2~2. 5, and relative reverse efficiency rates were 68% ~76% by Sibelium. Conclusion: Sibelium may re-verse the overexpression of MDR1.
