摘要
【目的】筛选桂枝甘草药对抗心律失常的有效部位。【方法】选用KM小鼠,随机分为空白对照组,模型组,维拉帕米组(剂量为0.1 g.kg-1.d-1),桂甘原液组、挥发油部位组、乙酸乙酯部位组、正丁醇部位组、母液部位组(剂量均为37 g.kg-1.d-1);采用经典水提法,系统溶剂萃取分离各部位,采用氯仿致小鼠心律失常动物模型,观察各组小鼠心律失常的发生率、死亡率、心率和心律失常持续时间。【结果】挥发油部位组和乙酸乙酯部位组小鼠的心律失常发生率显著低于模型组,死亡率显著低于维拉帕米组,且能显著减慢模型小鼠的心率,显著缩短模型小鼠的心律失常持续时间(均P<0.05)。【结论】挥发油部位和乙酸乙酯部位可能为桂枝甘草药对抗心律失常的有效部位。
Objective To screen the anti-arrhythmia effective components from coupled herbs of Ramulus Cinnamomi (RC) and Radix Glycyrrhizae (RG). Methods KM mice were randomized into blank control group, model group, verapamil (0. 1 g·kg^-1·d^-1 ) group, and the decoction, volatile oil fraction, acetoacetate fraction, n-butanol fraction, mother liquor of RC-RG groups (the dosage of all of them being 37 g ·kg^-1·d^-1 ). Classical water-extraction method and systemic solvent extraction method were used for the extraction of different fractions of coupled herbs of RC-RG. Mice models of arrhythmia were induced with chloroform. The incidence of arrhythmia, death rate, heart rate and duration of arrhythmia were observed in mice of different groups. Results In the groups of volatile oil fraction and acetoacetate fraction of RC-RG, the incidence of arrhythmia was lower, heart rate was slower, and arrhythmia duration was shorter than those in the model group, and the death rate was lower than that in verapamil group ( P 〈 0. 05 ). Conclusion It is suggested that volatile oil fraction and acetoacetate fraction be the effective fractions of coupled herbs of RC-RG.
